Feb. 17, 2011 - PRLog -- Tacrolimus (also FK-506 ) is an immunosuppressive drug that is mainly used after allogeneic organ transplant to reduce the activity of the patient's immune system and so lower the risk of organ rejection.

Tacrolimus ( FK506) is a macrolide lactone of fungal origin with strong immunosuppressive properties.It reduces interleukin-2 (IL-2) production by T-cells.

It has similar immunosuppressive properties to cyclosporin, but is much more potent in equal volumes. Immunosuppression with tacrolimus was associated with a significantly lower rate of acute rejection compared with cyclosporin-based immunosuppression Clinical outcome is better with tacrolimus than with cyclosporin during the first year of liver transplantation.

Tacrolimus is used to prevent organ rejection in liver transplant patients. It is also being used in experimental trials for kidney, heart, and bone marrow transplant patients.

Despite its therapeutic properties, tacrolimus exhibits some toxicity. Its adverse effects include nephrotoxicity, gastrointestinal tract complaints, neurotoxicity, and glucose intolerance.

Current immunoassays are practical for routine clinical use and allow for rapid analysis of tacrolimus in whole blood.
