A new software also allows for using the technology in regulated environments. A new kit for the fluorescent labeling of His-tagged proteins opens up unique possibilities for affinity analysis with MicroScale Thermophoresis (MST).

The automated nanoDSF workflows are fully integrated, single-vendor solutions that feature the following new instruments, consumables and software:

The new NanoTemper Technologies Prometheus NT.Plex is a high-performance nanoDSF instrument, that - same as the Prometheus NT.48 for single-capillary sample handling - provides high-quality protein unfolding curves, melting temperatures (Tm) and aggregation onsets (Tagg) when used with the new PR.ThermControl software for thermal unfolding experiments. In addition, both can provide Gibbs free enthalpy (ΔG) of a protein in just 3 seconds with the new PR.ChemControl software. The Prometheus NT.Plex supports the new capillary chips for nanoDSF, which enable automated workflows.

The Prometheus NT.Plex can be integrated with the vast majority of liquid handling platform using the new NanoTemper Technologies NT.Robotic Autosampler. The autosampler is a very flexible platform based on proven technology that does not only support automated sampling for up to 3 Prometheus NT.Plex, but can also be used with the NanoTemper Technologies Monolith NT.Automated or with combinations of both instruments. Depending on configuration, the number of samples that can be processed and analyzed in an automated fashion is virtually unlimited.

A strong demand for using nanoDSF technology in regulated environments is addressed by new software features that support GxP and 21CFR Part 11 compliance. The software is developed in close cooperation with partners in the pharmaceutical industry to ensure seamless integration in existing environments.

Instrument control, assay development and data evaluation reach new levels of simplicity with the new NanoTemper Technologies MO.Control software. The software leads the users intuitively through all steps of setting up a method and preparing the samples to run it. MO.Control even evaluates the data, finds possible mismatches and suggests ways to eliminate them. The software also includes a context-sensitive help and encyclopedia to answer possible questions right on the spot.

As an industry first, NanoTemper Technologies introduces the new Monolith NT His-Tag Labeling Kit RED-tris-NTA, which now enables straightforward labeling of His-tagged proteins for affinity analysis with MicroScale Thermophoresis. The label attaches non-covalently directly to the His-tag (His6 or larger) and requires no clean-up steps. This extremely simple and gentle labeling method will provide researchers with previously unknown insights in protein interactions at utmost data confidence.
