Giuseppe, MD, PhD, associate director for clinical research at Georgetown Lombardi Comprehensive Cancer Center，led the clinical trial of analyzing an individual's tumor to find and target genetic mutations that drive the cancer in order to assign cancer treatment.

The process went in a surprising fashion. "We expected it would take five years to enroll 600 patients into CUSTOM. But in less than two years, 668 patients were recruited," say Giuseppe.

The trial enrolled patients diagnosed with advanced stage non-small cell lung cancer, small cell lung cancer or thymic cancer. Then researchers use next-generation sequencing to look at their genes and assign them into different treatment groups based on genetic mutations or amplification. A new finding is that selumetinib, which is being studied for use in a number of cancers including non-small cell lung cancer, did not work on patients with KRAS mutations.

Although it is rather difficult to study cancer treatment for rare mutations because mutations can be in thousands of ways, this study shows that custom is going to be a new model for cancer treatment and drug development.
