Journal Article Highlights HemogloBind & AlbuVoid to Study Protein Organization in Red Blood Cells

The study defines a canonical RBC proteome and interactome using quantitative mass spectrometry (LC-MS) and machine learning. For this, the investigation analyzed and compared protein and RNA abundances (from MS and RNA-seq data) from RBCs, RBC precursor cells, other blood cell types (including WBCs, platelets, and reticulocytes) and plasma, with the derived likelihood score based solely on these measurements. The article states for red cell preparation, "For hemolysate preparations, remnant white ghosts were removed by centrifugation at 21,000 ×g for 40 min at 4°C, and the supernatant treated with HemogloBind (Biotech Support Group) in order to bind and remove free Hgb." For plasma preparation, "Plasma was centrifuged at 5,000 x g for 15 mins at 4°C, and only the top part of supernatant was collected. The supernatant was treated with AlbuVoid (Biotech Support Group) to reduce the presence of serum albumin (HSA)." Taken together, the study provides an in-depth view of the global protein organization of human RBCs and serves as a comprehensive resource for future research.
