MoBiTec GmbH Offers New Bacillus megaterium Strain for Gene Expression and Protein Production-YYBm1

MoBiTec GmbH expands its product line of tools for gene expression in Bacillus megaterium with the new expression host YYBm1. Strain YYBm1 is unable to utilize the expression system's inductor molecule xylose as carbon source.

GÖTTINGEN, Germany - Nov. 10, 2014 - PRLog -- Proteins are a group of organic molecules indispensable for life. Over the last decades, the intestinal Gram-negative bacterium Escherichia coli (commonly abbreviated E. coli) has been the preferred host for artificial gene expression. However, its Gram-negative status, giving cause to problems related to the presence of endotoxins, renders it unable to efficiently secrete heterologous proteins into the medium.

The Gram-positive bacterium Bacillus megaterium represents an increasingly used alternative for high yield intra- and extracellular protein production. During the past two decades, multiple tools including gene expression plasmids and production strains have been developed. Introduction of free replicating and integrative plasmids into B. megaterium is possible via protoplasts transformation or transconjugation. Using His- and Strep-affinity tags, the intra- or extracellular produced proteins can easily be purified in one-step procedures. Different gene expression systems based on the xylose controlled promoter P(xylA) and various phage RNA polymerase driven systems (for instance, T7) enable B. megaterium to produce up to 1.25 g of recombinant protein per liter.

MoBiTec GmbH is the only commercial source of gene expression systems for the biotechnologically relevant protein production host Bacillus megaterium.

As of now MoBiTec offers a new host strain for gene expression - YYBm1. Strain YYBm1 is unable to utilize the expression system's inductor molecule xylose as carbon source, thereby possibly making commercial applications  more efficient and less expensive. YYBm1 is supplied as ready-for-transformation protoplasts.
