June 25, 2012 - PRLog -- TACE Inhibitor Screening Assay Kit (Cat # K366-100): The TACE (tumor necrosis factor-α-converting enzyme), also called ADAM metallopeptidase domain 17 (ADAM17), is a 70-kDa enzyme that belongs to the ADAM protein family of disintegrins and metalloproteases. TACE is believed to be involved in the processing of tumor necrosis factor alpha (TNF-α) at the surface of the cell, and from within the intracellular membranes of the trans-Golgi network. This process, which is also known as 'shedding', involves the cleavage and release of a soluble ectodomain from membrane-bound pro-proteins (such as pro-TNF-α), and is of known physiological importance. In BioVision’s TACE inhibitor screening Kit, TACE hydrolyzes the specific FRET substrate to release the quenched fluorescent group, which can be detected fluorometrically. In the presence of the potent TACE inhibitor, the hydrolyzation of substrate will be impeded. Inhibitor Control GM6001 is included to compare the efficacy of test inhibitors.

Key features and benefits of the kit:

•Simple and convenient
•Rapid, reliable and sensitive
•Suitable for high-throughput
•Can be used for screening TACE inhibitors as well as detecting relative TACE activity
•Fluorometric quantification (Ex/Em = 318/449 nm)
•Applicable to various fields like metabolism, cell signaling, cancer, neurogenesis, apoptosis, Alzheimer's and inflammatory diseases.
•Kit provides ample reagents to perform 100 assays in a 96 well plate format

Peroxidase Activity Assay Kit (Cat # K772-100): Peroxidases (EC number 1.11.1.x) are a large family of enzymes that typically catalyze a reaction of the form: ROOR' + electron donor (2 e-) + 2H+ → ROH + R'OH. For many of these enzymes the optimal substrate is hydrogen peroxide, but others are more active with organic hydroperoxides such as lipid peroxides. Peroxidases can contain a heme cofactor in their active sites, or alternately redox-active cysteine or selenocysteine residues. BioVision’s Peroxidase Assay Kit provides a convenient colorimetric and fluorometric means to measure the peroxidase activity in biological samples. In the presence of Peroxidase, the OxiRed Probe reacts with Hydrogen peroxide in a 1:1 stoichiometry to produce the red-fluorescent oxidation product, resorufin. The resorufin is quantified by colorimetric ( λ= 570nm) or fluorometric methods (Ex/Em = 535/587 nm).

Key features and benefits of the kit:

•Simple and convenient
•Rapid, reliable and highly sensitive
•Suitable for high-throughput
•Versatile: can be used with plasma, serum, urine, cell culture supernatants or other biological fluids.
•Fluorometric (Ex/Em = 535/587 nm) or colorimetric (λ = 570 nm) quantification
•Can detect as little as 0.1 mU per well via colorimetric or 0.01 mU per well via fluorometric method, based on our unit definition.
•Applicable to various fields like oxidative stress, aging, neurodegeneration and autoimmune diseases.
•Kit provides ample reagents to perform 100 assays in a 96 well plate format
