GÖTTINGEN, Germany - June 5, 2014 - PRLog -- Western Blot is widely used in research to separate and identify proteins. In this technique a mixture of proteins is separated based on molecular weight through gel electrophoresis. The content of the gel is then transferred to a membrane producing a band for each protein. The protein of interest is then detected by means of a specific antibody.

Poly-histidine tags are often used for affinity purification of poly-histidine-tagged recombinant proteins expressed in Escherichia coli, Bacillus megaterium, Bacillus subtilis, and other prokaryotic expression systems. A poly-histidine tag is an amino acid motif in proteins that consists of multiple histidine (His) residues, mostly located at the N- or C-terminus of the protein. In order to facilitate the detection of successfully expressed his-tagged proteins the usage of anti-poly-histidine antibodies is a prevalent technique.

However, the common protocol for the antibody-based detection of the protein of interest is a laborious and time-consuming undertaking.
