Acetylcholine is an essential neurotransmitter, and inhibitors of cholinesterases(ChEs) are potent toxins. In a widely used enzymatic assay for evaluating ChE activity (Ellman et al), the presence of hemoglobin creates a substantial spectrophotometric background that interferes and must be overcome in order to accurately monitor cholinesterase activity from whole blood. In this article, a classical method was compared to a sample prep treatment incorporating HemogloBind™, Biotech Support Group’s unique solid-phase polymer for hemoglobin depletion. It was demonstrated that pretreatment of blood samples with Hemoglobind™ was a cost effective, high-throughput sample preparation alternative for obtaining total ChE activity and comparable to the classical method which involves separating plasma from red blood cells by centrifugation, lysing, and repeated washing of the cell membranes. With the latter classical method, preparing membranes prior to analysis is labor intensive and slow and removes plasma activity so total blood level activity cannot be efficiently assessed.
