There is a need to reassess this important class of drugs, and to reconcile the disconnect between the historic success of covalent drugs and the reluctance of most drug discovery teams to include them in their armamentarium. Dr. Singh’s presentation surveys the prevalence and pharmacologic features of FDA approved covalent drugs and describes specific examples of a new generation of rationally engineered covalent drugs and their potential in the treatment of cancer and infectious disease. The 5th Drug Design and Medicinal Chemistry Conference will discuss strategies for hit and target identification, fragment based drug design, chemoinformatics, protein-protein interactions, ligand based drug design, computer aided drug design, protein flexibility, high performance computing, covalent inhibitors, mutational resistance, selectivity of kinase inhibitors, potency, specificity, lead optimization, ADMET, safety in drug discovery, advances in drug delivery and much more.
